Description

Oral KPV capsules provide direct gut anti-inflammatory effects through intestinal MC1R activation. KPV (Lys-Pro-Val) is a tripeptide derived from the C-terminal sequence of alpha-melanocyte stimulating hormone (alpha-MSH), which is itself a potent endogenous anti-inflammatory neuropeptide. What makes KPV particularly unique among oral peptides is its ability to directly penetrate intestinal epithelial cells — most peptides cannot cross this barrier, but KPV’s small size and specific sequence allow it to enter cells directly and inhibit NF-kB signaling from within. Mechanism of Action

KPV activates melanocortin receptor 1 (MC1R) on intestinal epithelial cells and macrophages, triggering a signaling cascade that prevents NF-kB (nuclear factor kappa B) from translocating to the nucleus. NF-kB is the master regulator of inflammatory gene expression, and its inhibition reduces transcription of pro-inflammatory cytokines including IL-1beta, IL-6, TNF-alpha, and IL-8. Uniquely, KPV can also directly penetrate intestinal epithelial cells through a receptor-independent mechanism, allowing it to inhibit NF-kB signaling intracellularly. This dual mechanism — surface receptor activation and intracellular penetration — makes KPV more effective than compounds that can only act from the luminal surface. KPV also modulates macrophage polarization, shifting intestinal macrophages from a pro-inflammatory M1 phenotype toward an anti-inflammatory M2 phenotype, which promotes mucosal healing and reduces chronic inflammation.
