CJC-1295 No DAC binds to and activates GHRH receptors (GHRHR) on somatotroph cells in the anterior pituitary gland. GHRHR is a Gs protein-coupled receptor; its activation stimulates adenylyl cyclase, elevating intracellular cAMP, which activates protein kinase A (PKA) and ultimately drives GH synthesis and secretion. The short half-life ensures that pituitary stimulation occurs in discrete pulses rather than continuously, which is important because continuous GHRH stimulation leads to receptor downregulation and blunted GH response. When combined with Ipamorelin (a ghrelin receptor agonist), the two peptides act on different receptor systems — GHRHR and GHSR-1a respectively — producing a synergistic GH pulse that is 2–10 times greater than either agent alone. This combination also suppresses somatostatin (the endogenous GH inhibitor) through ghrelin receptor signaling, further amplifying the GH response.
