Description

5-Amino-1MQ is a small molecule inhibitor of NNMT (nicotinamide N-methyltransferase), an enzyme that depletes SAM (S-adenosylmethionine) and reduces NAD+ availability in metabolically active tissues. By inhibiting NNMT, 5-Amino-1MQ raises cellular NAD+ levels, activates AMPK, and promotes fat cell differentiation reversal — a process known as “whitening reversal” in adipose tissue. Research in obese mouse models demonstrated significant reductions in fat mass without changes in food intake, suggesting a direct metabolic effect rather than appetite suppression. The compound is notable for its ability to target visceral fat specifically, which is associated with metabolic syndrome and cardiovascular risk. As a small molecule, it is orally bioavailable and does not require injection, making it a convenient research tool for studying NAD+ metabolism and adipogenesis.

Mechanism of Action

“A small molecule NNMT inhibitor that promotes fat loss and metabolic health by raising cellular NAD+ levels.”

NNMT inhibition by 5-Amino-1MQ raises cellular NAD+ and SAM (S-adenosylmethionine) levels by preventing NNMT from consuming these substrates in the methylation of nicotinamide. Elevated NAD+ activates the sirtuin family of deacetylases (SIRT1–SIRT7), which regulate gene expression related to fat storage, mitochondrial biogenesis, and cellular aging. Simultaneously, elevated SAM supports epigenetic methylation reactions that promote metabolically favorable gene expression patterns. AMPK activation downstream of NAD+/SIRT1 signaling promotes fatty acid oxidation, inhibits lipogenesis, and enhances mitochondrial function. The net result is a shift in adipocyte biology from energy storage to energy expenditure, with demonstrated reversal of mature adipocyte differentiation back toward a pre-adipocyte phenotype in preclinical models.
