Description

MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino acid peptide encoded within the mitochondrial genome — specifically within the 12S rRNA gene. Discovered in 2015, it is the first mitochondria-derived peptide (MDP) shown to translocate to the nucleus and directly regulate nuclear gene expression, establishing a new class of mitochondria-to-nucleus communication. MOTS-c is considered a “mitokine” — a hormone-like signal released from mitochondria in response to metabolic stress. Circulating MOTS-c levels decline with age and correlate with metabolic health, and MOTS-c polymorphisms in humans are associated with longevity in Japanese centenarian studies. It has demonstrated remarkable effects on insulin sensitivity, exercise performance, metabolic homeostasis, and lifespan in multiple model organisms.
