Description

Methylene Blue (3,7-bis(dimethylamino)phenothiazin-5-ium chloride) is a synthetic compound with over 130 years of medical history, originally synthesized in 1876 and used as the first synthetic drug in medicine. It is FDA-approved for the treatment of methemoglobinemia and has been used as an antimalarial, antiseptic, and diagnostic agent. At low doses (0.5-4 mg/kg), Methylene Blue acts as an alternative electron carrier in the mitochondrial electron transport chain, improving ATP production efficiency and reducing the formation of reactive oxygen species. Multiple human studies have demonstrated cognitive enhancement effects, including improved memory consolidation, attention, and processing speed. Research has also shown potential neuroprotective effects relevant to Alzheimer’s disease, with Methylene Blue inhibiting tau aggregation and reducing amyloid toxicity. The compound’s unique redox cycling properties allow it to function as both an electron donor and acceptor, making it a versatile mitochondrial enhancer.
